TSLPisahemopoieticproteinthatisexpressedintheheart,liverandprostate.TSLPoverlapsBIOLOGicalactivitieswithIL-7,andbindswiththeheterodimericreceptorcomplexconsistingoftheIL-7Rα-chain(IL-7Rα)andtheTSLP-specificchain(TSLPR).LikeIL-7,TSLPinducesphosphorylationofSTAT3andSTAT5,butuseskinasesotherthantheJAKsforactivation.TSLPprohibitsapoptosisandstimulatesgrowthofthehumanacutemyeloidleukemia(AML)-derivedcelllineMUTZ3.ItinducesthereleaseofTcell-attractingchemokinesTARCandMDCfrommonocytes,andactivatesCD11c(+)dendriticcells(DCs).TSLP-activatedDCshavebeenshowntoprimenaïveTcellstoproducetheproallergiccytokines(IL-4,IL-5,IL-13,TNF-α)whiledown-regulatingIL-10 andIFN-γ,suggestingaroleininitiatingallergicinflammation.RecombinantHumanTSLPisa15.0 kDaproteinconsistingof132 aminoacidresidues.
