Description
Thrombomodulin(TM,CD141,THBD)isanendothelialcell-expressed,transmembraneglycoproteinthatcanformacomplexwiththecoagulationfactor,thrombin.Thethrombomodulin/thrombincomplexconvertsproteinCtoitsactivatedform,proteinCa,whichinturnproteolyticallycleavesanddeactivatesfactorVaandfactorVIIIa,twoessentialcomponentsofthecoagulationmechanism.Thisinactivationreducesthegenerationofadditionalthrombin,andtherebyeffectivelypreventscontinuedcoagulation.Reducedlevelsofthrombomodulincancorrelatewiththepathogenesisofcertaincardiovasculardiseases,suchasatherosclerosisandthrombosis.However,theserumlevelsofthetruncatedcirculatingformofthrombomodulinaretypicallyelevatedduringinflammationandinthepresenceofvariousinflammatory-relateddiseases.Thethrombomodulinproteincontains575 aminoacids,includingan18 a.a.signalsequence,a497 a.a.extracellulardomain,a24 a.a.transmembranesequence,anda36 a.a.cytoplasmicregion.RecombinantsolubleHumanThrombomodulinisa491 aminoacidglycoproteincontainingtheextracellulardomainofthrombomodulin.
