Description
Sox2belongstoadiversefamilyofstructurally-relatedtranscriptionfactorswhoseprimarystructurecontainsa79-residueDNA-bindingdomain,calledhighmobilitygroup(HMG)box.Itplaysanessentialroleinmaintainingthepluripotencyofembryonicstemcells(ESC)andthedeterminationofcellfate.MicroarrayanalysisshowedthatSox2regulatestheexpressionofmultiplegenesinvolvedinembryonicdevelopment,includingFGF-4,YES1andZFP206.Sox2actsasatranscriptionalactivatorafterformingaternarycomplexwithOct3/4andaconservednon-codingDNAsequence(CNS1)locatedapproximately2 kbupstreamoftheRAXpromoter.TheintroductionofSox2,Oct4,Myc,andKlf4intohumandermalfibroblastsisolatedfromaskinbiopsyofahealthyresearchfellowwassufficienttoconferapluripotentstateuponthefibroblastgenome.ThereprogrammedcellsthusobtainedresembleESCinmorphology,geneexpression,andintheircapacitytoformteratomasinimmune-deficientmice.Sox2andothertranscriptionfactorshavebeenintroducedintocellsbyDNAtransfection,viralinfection,ormicroinjection.ProteintransductionusingTATfusionproteinsrepresentsanalternativemethodologyforintroducingtranscriptionfactorsandothernuclearproteinsintoprimary,aswellastransformed,cells.RecombinantHumanSox2-TATexpressedinE.coliisa36.0 kDaproteincontaining330 aminoacidresidues,includingthe317 residuesoffull-lengthSox2anda13-residueC-terminalTATpeptide(GGYGRKKRRQRRR).
