TheIL-2 receptorsystemconsistsofthreenon-covalentlylinkedsubunitstermedIL-2Rα,IL-2Rβ,andIL-2Rγ.TheIL-2RαisatypeI transmembraneproteinconsistingofa219 aminoacidextracellulardomain,a19 aminoacidtransmembranedomainanda13 aminoacidintracellulardomain,whichisnotinvolvedinthetransductionofIL-2 signals.ProteolyticprocessingofIL-2RαreleasestheentireextracellulardomainofIL-2Rα,therebygeneratinga219 aminoacidsolubleproteincalledsolubleIL-2Rα(sIL-2Rα).ThehomodimericformbindsIL-2(KD=10mM)andfacilitatesIL-2 signaling.ThesecretedsIL-2Rαisexpressedonleukemiacells,lymphomacells,andnewlyactivatedTandBcells,aswellasonapproximately10%ofNKcells.RecombinantHumansIL-2Rαisa24.8 kDaproteincontaining219 aminoacidresiduesconsistingofonlytheextracellulardomainofIL-2Rα.Duetoglycosylation,IL-2Rαhasanapproximatemolecularweightof31 kDabasedonSDS-PAGEgelandMassSpectrometry.
