HumansDLL-4 comprisestheextracellularsignalingdomainofDLL,amemberofastructurally-relatedfamilyofsingle-passtypeI trans-membraneproteinsthatserveasligandsforNotchreceptors.DLL-4 functionstospecificallyactivatetheNotch-1 andNotch-4 receptors.TheNotchsignalingpathwayregulatesendothelialcelldifferentiation,proliferationandapoptosis,andisessentialforthedevelopment,maintenanceandremodelingofthevascularsystem.TargeteddeletionoftheDLL-4 geneinmiceresultedinseverevasculardefectsanddeathbeforebirth.Up-regulationofDLL-4 expressionhasbeenimplicatedinthevasculardevelopmentofcertaintumors.ThehumanDLL-4 geneconsistsofa503 aminoacidextracellulardomainwithoneDSLdomain,eightEGF-likerepeats,a21 a.a.transmembranedomain,anda135 a.a.cytoplasmicdomain.RecombinantHumansDLL-4 isa54.3 kDaglycoproteincontaining498 aminoacidresidues.
