HumansolubleDLL-1 comprisestheextracellularsignalingdomainofDLL-1,amemberoftheDelta/Serrate/Lag-2(DSL)familyofsingle-passtypeI trans-membraneproteinsthatserveasligandsforNotchreceptors.Itisexpressedprimarilyintheheart,pancreasandepidermis.DLL-1 functionstospecificallyactivatetheNotch-1 andNotch-2 receptors.ProteolyticcleavageofDLL-1 producesasecretedextracellulardomain,sDLL-1,thatinteractswithNotchreceptorsexpressedonadjacentcells.Notchsignalingplaysanessentialroleincontrollingcellfatedecisionsduringprenataldevelopmentandpostnatalstemcellrenewal,anddifferentiationinmanytissues.HumansDLL-1 blocksmonocytedifferentiationintomacrophages,butpermitsdifferentiationintodendriticcells.InhematopoieticProgenitorcells,hsDLL-1,suppressesdifferentiationintoB-cells,whilepromotingdifferentiationintoT-cellsandNKcellprecursors.Incellculture,humansDLL-1 hasbeenshowntopromoteexpansionofhematopoieticprogenitorcellsandsuppressapoptosisbyinhibitingdifferentiation.OverexpressionofNotchreceptorsappearstoinhibitdifferentiationinseveralmammaliancelllines,andincreasingevidencesuggeststhatNotchsignalingisfrequentlydownregulatedinhumanmalignancies.ThehumanDLL-1 geneconsistsofa528 aminoacidextracellulardomainwithoneDSLdomain,eightEGF-likerepeats,a23 aminoacidtransmembranedomain,anda155 aminoacidcytoplasmicdomain.ThecalculatedmolecularweightofRecombinantHumansDLL-1 is56.3 kDa.
