PAI-2 isaninhibitoryserpinexpressedmainlyinkeratinocytes,activatedmonocytes,andplacentaltrophoblasts.Itexistspredominantlyasa47 kDa,nonglycosylated,intracellularprotein,whichcanbeinducedtobesecretedas60 kDaglycoprotein.TheglycosylatedandunglycosylatedformsofPAI-2 areequallyeffectiveasinhibitorsofurokinase-typeplasminogenactivator(uPA),theonlyestablishedphysiologicaltargetofthisserpin.PAI-2 hasauniqueABIlitytoformdormantpolymersspontaneouslyandreversIBLyunderphysiologicalconditions.Thephysiologicalrelevanceofthisproperty,whichisneitheraconsequenceofanymutationinthePAI-2 genenorassociatedwithanyknowndisorder,isstillunclear.However,itappearsthattheformationofintracellular,dormantpolymersmaybeimportantforthecontrolledreleaseoftheinhibitorfromPAI-2 producingcells.PlasmalevelsofPAI-2 areusuallyloworundetectable,exceptduringpregnancyandinsomeformsofmonocyticleukemia.SecretionofPAI-2 fromtheplacentanormallyoccursduringthethirdtrimesterofpregnancy,andaccountsforthedramaticincreaseinPAI-2 levels(upto250 ng/ml),whicharemaintainedattheselevelsuntilpostpartum,andthenrapidlydecline.InadditiontoitsvitalroleinprotectingtheplacentafromdegradationbyuPAand/oruPA-activatedproteases,PAI-2 hasbeenshowntobeessentialforthepreventionofmetastaticspreadofneck,lungandbreastcancers.ThebeneficialeffectofPAI-2 seeninthesestudiesispresumedtostemfromitsabilitytoinhibituPA-dependentcelldissemination.PAI-2 hasalsobeenreportedtoinhibitkeratinocyteproliferation,andtoparticipateintheinnateimmuneresponseduringviralinfection.RecombinantHumanPAI-2 isa46.5 kDa,nonglycosylatedproteinof415 residues.
