Description

PlasminogenActivatorInhibitor-1(PAI-1,SerpinE1)isamemberoftheserpinfamilyofserineproteaseinhibitors,andistheprimaryinhibitorofurokinaseandtissueplasminogenactivator(tPA).PAI-1 isexpressedpredominantlyinADIpose,liverandvasculartissues,butisalsoproducedbycertaintumorcells.ElevatedlevelsofPAI-1 areassociatedwithobesity,diabetesandcardiovasculardisease,andincreasedproductionofPAI-1 isinducedbyvariousobesity-relatedfactors,suchasTNFα,glucose,insulin,andvery-low-densitylipoprotein.Theobesity-relatedelevationofPAI-1 levels,alongwiththeconsequentialdeficiencyinplasminogenactivators,canleaddirectlytoincreasedriskofthrombosisandothercoronarydiseases.Accordingly,PAI-1 hasbeenimplicatedasanimportantmolecularlinkbetweenobesityandcoronarydisease.PAI-1 canalsospecificallybindvitronectin(VTN)toformastableactivecomplexwithanincreasedcirculatoryhalf-liferelativetofreePAI-1. RecombinantHumanPAI-1 isa42.7 kDaproteincontaining379 aminoacidresidues.
