p16-INK4aisanuclearproteinthatregulatesthecellcyclebyinhibitingcyclin-dependentkinase-4(CDK4)andCDK6.p16-INK4ainhibitsCDKactivitybybindingtotheCDKmoleculesinamannerthatinterfereswiththeirABIlitytointeractwithcyclinD.Thisactivityhastheeffectofsuppressingtumorformationandgrowth,andofinducingreplicativesenescenceinvariousnormalcells,includingstemcells.Theexpressionofp16-INK4asteADIlyincreaseswithage,andtendstoaccumulateinstemcellcompartments.Thedeletion,rearrangement,ormutationofthep16-INK4ageneisfrequentlyfoundinmelanomas,aswellasincertainothertypesofcancer.p16-INK4aandothertranscriptionfactorshavebeenintroducedintocellsbyDNAtransfection,viralinfection,ormicroinjection.ProteintransductionusingTATfusionproteinsrepresentsanalternativemethodologyforintroducingtranscriptionfactorsandothernuclearproteinsintoprimary,aswellastransformed,cells.RecombinantHumanp16-INK4a-TATexpressedinE.coliisan18.0 kDaproteincontaining167 amino-acidresidues,includingthe155 residuesoffull-lengthp16-INK4aanda12-residueC-terminalTATpeptide(GYGRKKRRQRRR).
