p16-INK4aisanuclearproteinthatregulatesthecellcyclebyinhibitingcyclin-dependentkinase-4(CDK4)andCDK6.p16-INK4ainhibitsCDKactivitybybindingtotheCDKmoleculesinamannerthatinterfereswiththeirABIlitytointeractwithcyclinD.Thisactivityhastheeffectofsuppressingtumorformationandgrowth,andofinducingreplicativesenescenceinvariousnormalcells,includingstemcells.Theexpressionofp16-INK4asteADIlyincreaseswithage,andtendstoaccumulateinstemcellcompartments.Thedeletion,rearrangement,ormutationofthep16-INK4ageneisfrequentlyfoundinmelanomas,aswellasincertainothertypesofcancer.RecombinantHumanp16-INK4aisa16.5 kDaproteincontaining156 aminoacidresidues.Kim,W.Y.andSharpless,N.E.(2006)Cell127(2):265–75.
