NOVisamemberoftheCCNfamilyofsecreted,cysteine-richregulatoryproteins.ThefulllengthNOVproteincontainsfourstructuraldomainsthatconferdistinct,andsometimesopposing,BIOLOGicalactivities.ElevatedexpressionofNOVisassociatedwithcertaintumors,includingWilm’s tumorandmostnephroblastomas.However,inothertumortypesandcertaincancercelllines,increasedtumorgenicityandproliferationiscorrelatedwithdecreasedNOVexpression.Additionally,NOVinducescelladhesionandcellmigrationbysignalingthroughspecificcellsurfaceintegrins,andbybindingtoheparinsulfateproteoglycansandtofibulin1C.NOVhasalsobeenreportedtoexertproangiogenicactivities.RecombinantHumanNOVisa36.2 kDaproteincontaining331 aminoacidresidues.Itiscomposedoffourdistinctstructuraldomains(modules):theIGFbindingprotein(IGFBP)domain;thevonWillebrandFactorC(VWFC)domain;theThrombospondintype-I(TSPtype-1)domain;andaC-terminalcysteineknot-likedomain(CTCK).
