KLF4isamemberoftheKruppel-likefactor(KLF)familyofzincfingertranscriptionfactors.Membersofthisfamilyshare3 contiguousC2H2-typezincfingersatthecarboxylterminusthatcomprisetheDNA-bindingdomain.KLF4ishighlyexpressedinskinandgutepithelialtissues,butisalsofoundinvariousothercellsandtissues,includingvascularendothelialcells,lymphocytes,lung,andtestis.Itisanimportantregulatorofthecellcycle,transcription,andcelldifferentiation.TogetherwithSox2,Oct4,andcMyc,KLF4caninducethereprogrammingofprimaryhumanfibroblaststoapluripotentstate.KLF4andothertranscriptionfactorscanbeintroducedintocellsbyDNAtransfection,viralinfection,ormicroinjection.ProteintransductionusingTATfusionproteinsrepresentsanalternativemethodologyforintroducingtranscriptionfactorsintoprimary,aswellastransformed,cells.RecombinantHumanKLF4-TATisa51.7 kDaproteincontaining483 aminoacidresidues,includinga13-residueC-terminalTATpeptide.
