TheIGFsaremitogenic,polypeptidegrowthfactorsthatstimulatetheproliferationandsurvivalofvariouscelltypes,includingmuscle,bone,andcartilagetissueinvitro.IGFsarepredominantlyproducedbytheliver,althoughavarietyoftissuesproducetheIGFsatdistinctivetimes.TheIGFsbelongtotheInsulingenefamily,whichalsocontainsinsulinandrelaxin.TheIGFsaresimilartoinsulinbystructureandfunction,buthaveamuchhighergrowth-promotingactivitythaninsulin.IGF-IIexpressionisinfluencedbyplacentalactogen,whileIGF-I expressionisregulatedbygrowthhormone.BothIGF-I andIGF-IIsignalthroughthetyrosinekinasetypeI receptor(IGF-IR),butIGF-IIcanalsosignalthroughtheIGF-II/Mannose-6-phosphatereceptor.MatureIGFsaregeneratedbyproteolyticprocessingofinactiveprecursorproteins,whichcontainN-terminalandC-terminalpropeptideregions.RecombinantMurineIGF-I isa7.6 kDaglobularproteincontainingaminoacidsincluding3 intra-moleculardisulfide bonds.
