Description
TheIGFsaremitogenic,polypeptidegrowthfactorsthatstimulatetheproliferationandsurvivalofvariouscelltypes,includingmuscle,bone,andcartilagetissueinvitro.IGFsarepredominantlyproducedbytheliver,althoughavarietyoftissuesproducetheIGFsatdistinctivetimes.TheIGFsbelongtotheInsulingenefamily,whichalsocontainsinsulinandrelaxin.TheIGFsaresimilartoinsulinbystructureandfunction,buthaveamuchhighergrowth-promotingactivitythaninsulin.IGF-IIexpressionisinfluencedbyplacentalactogen,whileIGF-I expressionisregulatedbygrowthhormone.BothIGF-I andIGF-IIsignalthroughthetyrosinekinasetypeI receptor(IGF-IR),butIGF-IIcanalsosignalthroughtheIGF-II/Mannose-6-phosphatereceptor.MatureIGFsaregeneratedbyproteolyticprocessingofinactiveprecursorproteins,whichcontainN-terminalandC-terminalpropeptideregions.RecombinantHumanIGF-I andIGF-IIareglobularproteinscontaining70 and67 aminoacids,respectively,and3 intra-moleculardisulfidebonds.ThecalculatedmolecularweightofRecombinantHumanIGF-I is7.6 kDa.
