GDNFisadisulfide-linked,homodimericneurotrophicfactorstructurallyrelatedtoArtemin,NeurturinandPersephin.Theseproteinsbelongtothecysteine-knotsuperfamilyofgrowthfactorsthatassumestabledimericproteinstructures.GDNFsignalsthroughamulticomponentreceptorsystem,composedofaRETandoneofthefourGFRα(α1-α4)receptors.GDNFspecificallypromotesdopamineuptakeandsurvival,andmorphologicaldifferentiationofmidbrainneurons.UsingaParkinson’s diseasemousemodel,GDNFhasbeenshowntoimproveconditionssuchasbradykinesia,rigidity,andposturalinstABIlity.ThefunctionalratGDNFligandisadisulfide-linkedhomodimerconsistingoftwo15 kDapolypeptidechainscalledmonomers.Eachmonomercontainssevenconservedcysteineresidues,includingCys-101,whichisusedforinter-chaindisulfidebridging,andothersthatareinvolvedintheintramolecularringformationknownasthecysteineknotconfiguration.ThecalculatedmolecularweightofRecombinantRatGDNFis30.0 kDa.
