GDF-15 belongstotheTGF-βcytokinefamily,whosemembersplayanimportantroleduringprenataldevelopmentandpostnatalgrowth,andtheremodelingandmaintenanceofavarietyoftissuesandorgans.GDF-15 isexpressedpredominantlyintheplacentaand,toamuchlesserextent,invariousothertissues.ThepresenceofGDF-15 inamnioticfluidanditselevatedlevelsintheseraofpregnantwomensuggestGDF-15"s involvementingestationandembryonicdevelopment.GDF-15 generallyexertstumorsuppressiveactivitiesandisoneofthepredominantfactorsproducedandsecretedinresponsetoactivationofthep53pathway.Interestingly,theserumlevelofGDF-15 ispositivelycorrelatedwithneoplasticprogressionofseveraltumortypes,includingcertaincolorectal,pancreatic,andprostatecancers.HumanGDF-15/MIC-1 isadisulfidelinkedhomodimericproteinconsistingoftwo112 aminoacidpolypeptidechains.ThecalculatedmolecularweightofHumanGDF-15/MIC-1 is24.6 kDa.
