Selectinsareafamilyofcalcium-dependenttype1 transmembraneproteins.Endothelial(E)-selectinisaheavilyglycosylatedtransmembraneproteinexpressedbyactivatedendothelialcellsinmicrovascularlinings.E-selectin,alongwithP-selectinandL-selectin,initiaterecruitmentofcirculatingleukocytesfrombloodtositesofinflammationinthevascularliningthroughinteractionwithspecificcellsurface-associatedcarbohydratedeterminants.E-selectinconsistsofanN-terminaltype1 lectindomain,anEGF-likedomain,6 sushi(CCP/SCR)domains,atransmembranesequence,andashortcytoplasmicdomain.RecombinantHumanE-selectinisa58.6 kDaproteincontaining535 aminoacidresidues,correspondingtotheextracellularportionofthefulllengthprotein.Duetoglycosylation,E-selectinmigratesatanapparentmolecularweightofapproximately65–85kDabySDS-PAGEanalysisunderreducingconditions.
