DKK-1 isamemberoftheDKKproteinfamilywhichalsoincludesDKK-2,DKK-3 andDKK-4. DKK-1 wasoriginallyidentifiedasaXenopusheadformingmoleculethatbehavesasanantagoNISTforWntsignaling. SubsequentstudieshaveshownthatDKK-1 andDKK-4 playanimportantregulatoryroleintheWnt/β-cateninsignalingpathwaybyforminginhibitorycomplexeswithLDLreceptor-relatedproteins5 and6(LRP5andLRP6),whichareessentialcomponentsoftheWnt/β-cateninsignalingsystem.LPR5andLPR6aresingle-passtransmembraneproteinsthatappeartoactasco-receptorsforWntligandsinvolvedintheWnt/β-cateninsignalingcascade.IthasbeensuggestedthatbyinhibitingWnt/β-cateninsignaling,whichisessentialforposteriorpatterninginvertebrates,DKK-1 permitsanteriordevelopment.ThisnotionissupportedbythefindingthatmicedeficientofDKK-1 expressionlackheadformationanddieduringembryogenesis.RecombinanthumanDKK-1 expressedinhuman293 cellsisa35–40kDaglycoproteincontaining235 amino-acidresidues.
