Description
TGF-βfamilymembersarekeymodulatorsofcellproliferation,differentiation,matrixsynthesis andapoptosis.Asimpliedbytheirname,BMPsinitiate,promote andregulatethedevelopment,growthandremodelingofboneandcartilage.Inadditiontothisrole,BMPsarealsoinvolvedinprenataldevelopmentandpostnatalgrowth,remodelingandmaintenanceofavarietyofothertissuesandorgans.BMP-7,alsoknownasosteogenicprotein-1 orOP-1,isapotentboneinducingagent,whichinthepresenceofappropriateosteoconductivecarrier(e.g.collagenspongeorsynthetichydroxyapatite)canbeusedinthetreatmentofbonedefects.A bone-graftsubstitute,calledOP-1™implant,madeofrecombinanthumanBMP-7 associatedwithbovinebone-derivedcollagen,hasrecentlybeenapprovedbytheFDAasadevicefortreatingcritical-sizebonefractures.ThepotentialuseofBMP-7 indentalreconstructivesurgeriesiscurrentlyunderinvestigation.RecombinanthumanBMP-7 isa28.8 kDahomodimericglycoproteinconsistingoftwo117 aminoacidsubunits,whichcorrespondtoaminoacidresidues315 to431 ofthefull-lengthBMP-7 precursor.
