ApoA-I isa29.0 kDaproteinproducedintheliverandintestine,andsecretedasthepredominantconstituentofnascenthighdensitylipoprotein(HDL)particle.ApoA-I,whichisfoundexclusivelyinHDL,hasauniqueABIlitytocaptureandsolubilizefreecholesterol.ThisApoA-I abilityenablesHDLtoremoveexcessperipheralcholesterol,andreturnittotheliverforrecyclingandexcretion.Thisprocess,calledreversecholesteroltransport,isthoughttoinhibitatherogenesis.Forthisreason,HDLisalsoknownasthe“goodcholesterol.”ThetherapeuticpotentialofApoA-I hasbeenrecentlyassessedinpatientswithacutecoronarysyndromes,usingarecombinantformofanaturallyoccurringvariantofApoA-I(calledApoA-I Milano).TheavailabilityofrecombinantnormalApoA-I shouldfacilitatefurtherinvestigationintothepotentialusefulnessofApoA-I inpreventingatheroscleroticvasculardiseases.RecombinantHumanApoA-I isa28.2 kDaproteinof244 aminoacidresidues.
