ANG-2 bindstotheendothelialcellspecificreceptorTie-2,but,incontrasttoANG-1,doesnotinducetyrosinephosphorylation.Consequently,ANG-2 modulatesANG-1 activationofTie-2 and,dependingonthephysiologicalandbiochemicalenvironment,canacteitherasanagoNISTorantagonistofTie-2 inducedangiogenesis.ThesignalinginteractionsofANG-1,ANG-2 andTie-2,alongwithlesscharacterizedANG-3 andANG-4,arerequiredforembryonicandadultangiogenesis.Physiologically,ANG-1 andANG-2 areassociatedwithsprouting,tubeformation,andstructuralintegrityofnewlyformedbloodvessels.MaturehumanANG-2 isasecretedproteincontaining480 aminoacidresidues.ANG-2 iscomposedofanalpha-helix-rich“coiledcoil”N-terminaldomainandfibrinogen-likeC-terminaldomain.ANG-2 existspredominantlyintheformofadisulfide-linkeddimer.RecombinantHumanANG-2 isaC-terminalhistidine-taggedglycoproteinwhichmigrateswithanapparentmolecularmassof60.0 –70.0 kDabySDS-PAGEunderreducingconditions.SequencinganalysisshowsanN-terminalsequencestartingwithresidue68(D)oftheANG-2 precursorprotein.ThecalculatedmolecularweightofRecombinantHumanANG-2 is50.1 kDa.
