Description
Angiopoietin-1(Ang-1)isasecretedligandforTie-2,atyrosine-kinasereceptorexpressedprimarilyonvascularendothelialcellsandearlyhematopoieticcells.Ang-1/Tie-2 signalingpromotesangiogenesisduringthedevelopment,remodeling,andrepairofthevascularsystem.TransgenicmicelackingexpressionofeitherAng-1 orTie-2 failtodevelopafullyfunctionalcardiovascularsystemanddiebeforebirth.Postnatally,theangiogenicactivityofAng-1/Tie-2 isrequiredduringnormaltissuerepairandremodelingofthefemaleendometriuminthemenstrualcycle.Ang-1/Tie-2 signalingappearstoberegulatedbyAngiopoietin-2(Ang-2),anaturalantagoNISTforTie-2 thatexertsitseffectsthroughaninternalautocrineloopmechanism.Inadditiontosuppressingendothelialcellactivationbyinhibitingtheexpressionofadhesionandinflammatorymolecules,Ang-1 enhancesendothelialcellsurvivalandcapillarymorphogenesis,andlessenscapillarypermeABIlity.Assuch,Ang-1 haspotentialtobecomeaneffectivetherapeuticagentfortreatingvariousendotheliumdisorders,includingseveralseverehumanpulmonarydiseases.Theefficacyofcell-basedAng-1 genetherapyforacutelunginjury(ALI)hasrecentlybeenstudiedinaratmodelofALI.TheresultsofthisstudyshowthatsuchtherapycanmarkedlyimprovelungconditionandsuggestthatAng-1 therapymayrepresentapotentialnewstrategyforthetreatmentand/orpreventionofacuterespiratorydistressinjury(ARDI),asignificantcauseofmorbidityandmortalityincriticallyillpatients.RecombinanthumanANG-1,derivedfromHeLacells,isaC-terminalhistidinetaggedglycoproteinwhichmigrateswithanapparentmolecularmassof60.0 –70.0 kDabySDS-PAGEunderreducingconditions.SequencinganalysisshowsN-terminalsequencesstartingwithSer-20 andwithAsp-70 ofthe498 aminoacidprecursorprotein.ThecalculatedmolecularweightofRecombinantHumanANG-1 is56.3 kDa.
